Product Notes
- AciI has a non-palindromic recognition site. As a result, when DNA is cut with AciI and then ligated, only 50% of these ligated sites regenerate an AciI site.
- This enzyme does not benefit from incubations over 1 hour. Visit "Restriction Endonucleases-Survival in a Reaction" for more information.
- For enzymes that cannot be heat-inactivated, we recommend using a column for cleanup (such as the Monarch® PCR & DNA Cleanup Kit), or running the reaction on an agarose gel and then extracting the DNA (we recommend Monarch Gel Extraction Kit), or performing a phenol/chloroform extraction.
- Blocked by CpG methylation.

FAQs & Troubleshooting
FAQs
- Why can only 50% of AciI cut and ligated fragments be recleaved by AciI?
- Do I have to set-up digests with Time-Saver™ qualified enzymes for 5-15 minutes? Can I digest longer?
- Is this enzyme sensitive to dam, dcm or mammalian CpG methylation?
- Which NEB restriction enzymes are supplied with Gel Loading Dye, Purple (6X)?
- Can you tell me more about the switch from BSA to Recombinant Albumin (rAlbumin) in NEBuffers?
Tech Tips
This enzyme typically leaves very small DNA fragments, so a 2% agarose gel is suggested for separating the small fragments.
An over digest may be needed to cleave PCR fragments, the TAQ Pol reaction buffer and primers from the PCR reaction tend to inhibit Aci I.
