Description

The Monarch StabiLyse DNA/RNA Buffer is a dual-function reagent for nucleic acid stabilization and sample lysis. This proprietary formulation preserves the integrity of DNA and RNA in samples during storage and it also enables effective sample lysis from a wide variety of eukaryotic and prokaryotic sample types. Samples can be stored in the Monarch StabiLyse DNA/RNA Buffer at the point of harvest for short and long term without the need for immediate processing. The Monarch StabiLyse DNA/RNA Buffer is unique in that it does not require removal of this reagent or sample precipitation.

The Monarch StabiLyse DNA/RNA Buffer is ideal for performing sample homogenization and mechanical disruption. The extracted nucleic acids are suitable for a variety of downstream applications including PCR, sequencing, and more.

10 mg pieces of rat liver were stored in 100 µl Monarch StabiLyse DNA/RNA Buffer at the time of harvest and placed at room temperature, 4°C and –80°C. DNA extraction was performed using Monarch® Spin gDNA Extraction Kit (NEB #T3010) following the tissue protocol. Isolated gDNA was run on Genomic DNA ScreenTape® on Agilent® TapeStation® 4200 (Agilent Technologies). High quality DNA (> 8.0) was observed after sample storage for 1 week at room temperature, 4 weeks at 4°C and 4 weeks at –80°C.

RNA extraction was performed using Monarch Spin RNA Isolation Kit (Mini) (NEB #T2110) following the tissue protocol. Isolated RNA was run on RNA ScreenTape on Agilent TapeStation 4200 (Agilent Technologies). High quality RNA (> 8.0) was observed after sample storage for 1 week at room temperature, 4 weeks at 4°C and 4 weeks at –80°C.

Pellets of 1 million HeLa cells were resuspended in either Monarch StabiLyse DNA/RNA Buffer or Phosphate-buffered saline (PBS) and incubated at 37°C for 3 hours. RNA extraction was performed using Monarch Spin RNA Isolation Kit (Mini) (NEB #T2110). Isolated RNA was run on RNA ScreenTape on Agilent TapeStation 4200 (Agilent Technologies). Cells resuspended in Monarch StabiLyse DNA/RNA Buffer showed intact RNA compared to PBS after heat exposure indicating protection against RNA degradation.
