- Based on the stability of the enzyme in the reaction, incubations longer than 1 hr will not result in improved digestion, unless additional enzyme is added. Please refer to Restriction endonuclease survival in a reaction for more information regarding this topic.
- NEB sells two neoschizomers of KasI. KasI cleaves G/GCGCC, NarI cuts GG/CGCC and SfoI cuts to leave blunt GGC/GCC ends. Both KasI and NarI have sitepreference, cleaving some sites very slowly. SfoI does not show sitepreference.
- Blocked by CpG methylation.
- star activity may result from a glycerol concentration of >5%

- Is KasI affected by methylation?
- Is extended digestion of KasI recommended?
