Principle / Procedure
NucleoMag® DNA/RNA Water enables manual or automated detection of either DNA, RNA, or both molecules in parallel from a range of water samples, spanning from turbid to clear. NucleoMag® DNA/RNA is compatible with a variety of filters and filtration systems, including 25 mm and 47 mm filters from various materials as well as with cartridge filters (such as Sterivex™). This makes NucleoMag® DNA/RNA Water ideal for microbial metagenomics, environmental microbiology as well as for estimation of water quality through determination of the microbial load of the sample. As DNA is extracted from the samples, presence of the entire microbiome, including coliform, pathogenic, as well as free living water microorganisms can be measured directly. The concurrent isolation of DNA and RNA allows for determination of the amount of living microorganisms in the sample and estimating, e.g., levels of gene expression as well as efficiencies of sterilization procedures. NucleoMag® DNA/RNA Water comes with magnetic beads and buffers included. The kit can additionally be combined with RNase, DNase, and two sizes of NucleoSpin® Bead Tubes Type A, providing a range of different options for the choice of filter and nucleic acid. In addition to manual nucleic acid purification, NucleoMag® DNA/RNA Water protocols are automation friendly and compatible with automation platforms suitable for magnetic bead purifications, such as KingFisher® and epMotion®. The kit does not require the addition of any reducing agents.

Application data
Efficient detection for different water and air samples
Various water samples and an air sample were filtered and the extracted DNA was analyzed by PCR. Microbial DNA could be efficiently measured for all of the samples, demonstrating the versatility of the NucleoMag® DNA/RNA Water kit.
Compatibility with different homogenization devices
A lysis reaction was performed with homogenization on either a Vortex-Genie® 2 device or a dedicated homogenization device. Both approaches delivered comparable results indicating the flexibility of the NucleoMag®DNA/RNA Water kit with regards of homogenization platforms.
Compatibility with different filtration systems
A qPCR was performed with nucleic acids isolated from round filters and a filtration cartridge system, demonstrating reliable results across different Filtration systems.



No PCR inhibition
Serial dilutions were performed for three different water samples. The linearity of the inhibition plots indicates the absence of PCR inhibition.
Automated protocol
In addition to manual processing, NucleoMag® DNA/RNA Water allows for automated workflows. Here, two different water samples were processed on two different automated platforms- a liquid handler and a magnetic rod system. Both systems delivered comparable results, indicating the adaptability of the kit to both automation approaches.
