Purification of highly concentrated RNA from smallest samples

NucleoSpin® RNA XS allows the isolation of highly concentrated and highly pure RNA from small samples. It is based on the reliable silica membrane technology, featuring the innovative NucleoSpin® XS column design which allows elution in only 5–30 µL resulting in high RNA concentrations. For isolation of high-quality RNA it is important to prevent degradation of the RNA and to eliminate genomic DNA. With the NucleoSpin® RNA XS method, cells are lysed by incubation in a solution containing large amounts of chaotropic ions (see diagram "NucleoSpin® RNA XS procedure"). This lysis buffer immediately inactivates RNases – which are present in virtually all biological materials – and creates appropriate binding conditions which favour adsorption of RNA to the silica membrane. After lysis, homogenization and reduction of viscosity are achieved by filtration with NucleoSpin® Filter units. Residual genomic DNA is removed efficiently by time-saving on-column digestion with rDNase, provided with the kit. Total RNA is eluted in RNAse free water. The low elution volume of only 5–30 µL leads to concentrated RNA with typical RIN (RNA Integrity Numbers) > 9 (depending on sample quality). NucleoSpin® RNA XS is the ideal solution for smallest samples like microdissected tissues, minute amounts of plant material, or even from a single cell.

The innovative funnel shaped thrust ring of NucleoSpin® RNA XS holds a silica membrane of very small diameter. Thus very efficient elution in only 5–30 μL can be achieved. Hence the NucleoSpin® RNA XS is the ideal solution for extraction of RNA from trace amounts of sample giving best results in sensitive downstream applications.
