One unit is defined as the amount of enzyme required to digest 1 µg of λ DNA in 1 hour at 37°C in a total reaction volume of 50 µl.

  1. NEB sells two neoschizomers of PleI. PleI leaves a 5´ overhang, MlyI is a blunt cutter while Nt.BstNBI is a nicking enzyme.

  3. Blocked by some combinations of overlapping CpG methylation.

  1. Is PleI activity sensitive to dam, dcm or mammalian CpG methylation?
  2. How many base pairs should be added at the end of a PCR primer next to the PleI recognition site to guarantee that PleI will cut properly?
