Principle

The NucleoMag® DNA Bacteria kit has been developed for high throughput, automation friendly isolation of genomic DNA from diverse microbial cultures, such as Gram-negative and Gram-positive bacteria as well as yeasts.
NucleoMag® DNA Bacteria utilizes a powerful yet environmentally friendly buffer chemistry, free of chaotropic salts (patent pending). The kit is free from any dangerous goods, including highly concentrated alcohols, enabling simpler and less expensive transport as well as more convenient handling and waste disposal.
For optimal DNA yields, a complete disruption of sample material is necessary. For disruption of bacterial cell pellets, we recommend combining the kit with MN Bead Tubes Type B (40-400 µm glass beads), for lysis of yeast cells we recommend MN Bead Tubes Type A (0.6-0.8 mm ceramic beads) while insect samples, and lipid tissues and fungi should be homogenized with MN Bead Tubes Type D (3 mm steel beads). MN Bead Plate Type B is loaded with 40-400 µm glass beads and is intended for homogenization of bacterial samples while the MN Bead Plate Type D is loaded with 3 mm steel balls and is optimized for homogenization of insects, crustaceans, lipid tissues and fungal samples.
After sample disruption, Binding Buffer IMB and the NucleoMag® B-Beads are added to the transferred lysate. Subsequent to the magnetic separation, the NucleoMag® B-Beads are washed to remove contaminants and salts using Wash Buffer IMW and 80 % ethanol (600 μL each) respectively. After air drying the NucleoMag® B-Beads for 10 min at RT, the DNA is finally eluted with Elution Buffer IME (50–200 μL).
The NucleoMag® DNA Bacteria kit can be used either manually or automated on standard liquid handling instruments and automated magnetic separators.
