Principle
Isolation of genomic DNA from soil and sediments
The NucleoSpin® 8 / 96 Soil kits allow manual and automated purification of DNA from soil samples using proven silica-membrane technology. The kit includes two alternative lysis buffers and an additional Enhancer SX. The option of two buffers and an enhancer allows tuning the optimal lysis conditions for highest yield from different soil samples of different compositions. Soil samples exhibit different pHs, contain different amounts of organic or anorganic matter, such as humic substances or polysaccharides. The lysis options and the unique NucleoSpin® Inhibitor Removal Strip/Plate provide a removal of all PCR inhibitors during the purification procedure. The NucleoSpin® Bead Tubes, containing ceramic beads, allow a most efficient lysis of microorganisms in the sample. The maximum amount of 500 mg soil as starting material yields typically 2- 10 µg DNA. The eluted genomic DNA is ready to use for downstream applications, such as PCR, qPCR, restrictions analysis, and others. Diluting the eluated DNA is not necessary. The NucleoSpin® 8 / 96 Soil kits are for manual and automated use.
