Product Introduction

The blue protein loading dye contains one vial of blue loading buffer and one vial of 30X reducing agent. The combined solution is ideal for protein gel applications.

  • 1X Blue Loading Buffer Composition: 62.5 mM Tris-HCl (pH 6.8), 2% (w/v) SDS, 10% glycerol, 0.01% (w/v) bromophenol blue
  • 30X Reducing Agent: 1.25 M DTT
  • SDS interacts with positively charged amino acids of proteins, thereby disrupting interactions that form protein structures to separate proteins based on size rather than charge or shape 
  • DTT is present to help reduce structural disulfide bonds

Properties & Usage

Product Notes

  1. Reagents stored at different temperatures.
  2. Prepare fresh 3X Reducing Blue Loading Buffer by adding 1/10 volume 30X Reducing Agent to 1 volume of 3X Blue Loading Buffer.
  3. Prepare samples by adding 2 volumes of Sample to 1 volume 3X Reducing Blue Loading Buffer (from step 1).
  4. Heat samples to 95-100°C for 3-5 minutes.
  5. After a quick microcentrifuge spin, load directly on to a gel. To ensure uniform mobiity, load an equal volume of 1X Reducing Blue Loading Buffer into any unused wells.

DTT will degrade at room temperature. Keep the two components separate at store at their proper temps.
