One unit is defined as the amount of enzyme required to digest 1 µg of λ DNA (dam-) in 1 hour at 37°C in a total reaction volume of 50 µl.

Product Notes

  1. BfuCI, DpnII and Sau3AI are isoschizomers of MboI.
  2. MboI cleaves to leave a 5´ GATC extension which can be efficiently ligated into BamHI, BclI, BglII, BstYI, DpnII or Sau3AI cleaved fragments.
  3. MboI is blocked by dam methylation, however Sau3AI is not sensitive to dam methylation.
  4. This enzyme is blocked by dam methylation. More information can be found at Dam-Dcm and CpG Methylation.
  5. Based on the stability of the enzyme in the reaction, incubations longer than 1 hr will not result in improved digestion, unless additional enzyme is added. Please refer to Restriction endonuclease survival in a reaction for more information regarding this topic.
  6. Blocked by dam methylation and impaired by overlapping CpG methylation.

Troubleshooting

Tech Tips

This enzyme will not cut plasmids isolated from most E.coli because it is blocked by dam methylation. For enzyme cleavage to occur, plasmids must be transformed and isolated from a dam-, dcm- strain of E.coli, such as NEB’s dam-/dcm- Competent E.coli (NEB #C2925).
